1. To ensure the accuracy of the pipette gun, the error cannot exceed 2%. Can be determined with water and electronic balance. But it is best to have a professional correct it.
2. To be equipped with 20ul, 50ul, 100ul, 1000ul and a gun each. After aspirating a different liquid, replace the pipette tip. Even when aspirating standards.
3. Take the kit out of the refrigerator 1 hour before the experiment, and let all the reagents return to room temperature to make the results more stable.
4. During the experiment, keep the substrate away from light.
5. When using the gun to absorb the liquid, the speed should not be too fast, so as to avoid bubbles and inaccurate suction.
6. When sucking liquid, use a gun with a range close to the required amount to reduce errors.
7. When adding the liquid into the enzyme-labeled well, avoid the contact between the pipette tip and the liquid in the hole, so that the droplets on the pipette tip can contact the hole wall, and the droplets will flow down naturally.
8. After all the liquid is added, gently shake the ELISA plate on the table for 30 seconds in parallel to mix the liquid. The shaking function of the microplate reader can also be used.
9. When taking a warm bath, seal the ELISA plate with self-adhesive or tape paper to prevent the evaporation of water.
10. When washing the plate, after each addition of the lotion, it should be left for 1 minute to make the cleaning more thorough. When a plate washer is not available, pat the plate dry on newspaper or wool paper after removing the liquid.
11. When the washing liquid is not enough, you can use distilled water to prepare PH7.4, 0.02M phosphate buffer, and add 0.1% Tween 20 as the washing liquid. It can be stored for a long time after adding 1/1000 of sodium azide.
12. The substrate is light-sensitive and should be prepared immediately before use.
13. Before testing, turn on the microplate reader and make it stable for more than 10 minutes.
14. The substrate has certain toxicity, and the stop solution is corrosive to the skin, so it should be avoided as much as possible.
15. The sample to be tested should be clarified, otherwise it will affect the result.
16. The warm bath time should comply with the kit regulations.
17. Double-hole experiments should be done as much as possible, so as to ensure the accuracy of the data.
18. Samples with questionable results should be confirmed by other methods.
